چکیده

The aim of this novel endoscopy clinic-based case-control study was to explore the influence of different Helicobacter pylori strain types on the risk of gastric adenocarcinoma using isolated bacterial strains, tissue samples, and sera. We included 72 cases with gastric adenocarcinoma and 324 ageand sex-matched controls. Histological characterization, culture, molecular typing of H. pylori genes by PCR (cagA/ vacA), conventional IgG ELISA, and immunoblotting (Western blot) for the CagA and VacA proteins were performed. With four tests combined, H. pylori infection was detected in 57 (79%) cases and 213 (66%) controls. A positive association between H. pylori infection and gastric cancer risk was found [odds ratio (OR), 2.1; 95% confidence interval, 1.1–3.9]. Type I (OR, 1.8), intermediate (OR, 2.0), and type II (OR, 0.2) strains of H. pylori presented different serum antibody levels and different levels of association with gastric cancer. Our case-control study, based on molecular characterization and serology, provides further evidence that infection by more virulent strains of H. pylori and the presence of antibodies toward the CagA protein can be used as markers for an increased risk of gastric adenocarcinoma and that the strain types of H. pylori could be used in the future to determine disease outcome.

The Role of Interleukin 1β Gene Polymorphism in Gastric Cancer in North-eastern Iran

Background and Objective: Host genetic factors such as cytokine gene polymorphisms as well as Helicobacter pylori (H. pylori) infection have been found to be associated with gastric cancer risk . Interleukin 1 is a pro-inflammatory cytokine involved in H. pylori-induced gastric inflammation. Therefore, w...

Prevalence of Helicobacter Pylori Infection in Asymptomatic Children in Birjand, Eastern Iran

Background: Helicobacter pylori is the cause of serious diseases including gastric cancer and gastric mucosa–associated lymphoid tissue lymphoma.50% of world population is infected by this microorganism and it -based on epidemiologic studies - is mainly acquired during childhood . there is not enough evidence about prevalence of this infection in children and its risk factors so encourage us to...

Impact of Methylenetetrahydrofolate Reductase C677T Polymorphism on the Risk of Gastric Cancer and Its Interaction with Helicobacter pylori Infection

Background: Attempts for early detection of gastric cancer have recently focused on host's genetic susceptibility factors and gene-environment interactions. We have, herein, studied the association of MTHFR C677T single nucleotide polymorphism (SNP) and its interaction with Helicobacter pylori infection, smoking, age and gender on the risk of gastric cancer among an Iranian population. Methods:...

Analysis of KRAS gene mutation associated with Helicobacter pylori infection in patients with gastric cancer

Objective(s): KRAS proto-oncogene mutation can be considered a diagnostic factor for treating various malignancies. Helicobacter pylori infection, a risk factor for stomach cancer, may cause DNA damage and genetic changes. The aim of the current study was to assess the association of gastric cancer and KRAS mutation, demographic factors, and H. pylori infection.<...
